Overview

MOTS-C is a mitochondrial-derived peptide encoded directly in mitochondrial DNA — making it unique among peptides in that it originates outside the nuclear genome entirely. It functions as a signalling molecule communicating metabolic stress from the mitochondria to the rest of the cell, triggering adaptive responses that improve energy efficiency, insulin sensitivity, and stress resilience. It is frequently described in research literature as an “exercise mimetic” — activating many of the same metabolic pathways as physical exercise, without providing the mechanical stimulus for muscle growth.

 

What it does

MOTS-C production increases in response to low glucose, exercise, oxidative stress, and mitochondrial dysfunction. It activates AMPK — the master metabolic regulator — improving mitochondrial health, enhancing fat and glucose utilisation, increasing insulin sensitivity, and promoting metabolic flexibility. In aging research, it counteracts age-related mitochondrial decline by improving mitochondrial signalling and energy production. It has been described in longevity research as one of the most promising mitochondrially-encoded regulators of biological aging.
